Here's the short version: The website/youtube-skit-channel "College Humor" made it's own video streaming platform called Dropout. Game Changer is a Gameshow on dropout wherein the rules and nature of the game change every episode, and the contestants are not told what the rules of the game are.
Part of me thinks this must be scripted because it's so perfect, but another part of me realizes that if anyone could ad-lib something like this, it's Brennan Lee Mulligan.
Also, Zac's "whoa, dude" is the cherry on top.
Brennan Lee Mulligan has said on Twitter (about this clip) that it takes actual effort for him to just not speak like this all the time.
It's going to be such a bittersweet moment when Brennan eventually leaves CollegeHumor/Dropout for bigger and better things. He, Sam, and Trapp form this wonderful trifecta.
He basically *is* collegehumor at this point

As someone in the YouTube comments put it, they pitted the single most competitive cast member against the two most chill cast members and it was perfect.

And for people who watch Critical Role but are yet to give Dimension20 a chance - Matt Mercer is one of the PCs in that one. It's only 6 episodes, really do recommend.
Some context -- Game Changer is a game show where the rules change every show, and the players need to figure out the rules as they go. This show the players were just asked "Yes or No" in various ways, with the goal of figuring out the rule. The players were musing out loud if the answer was determined by the way the question was posed, the answers of their other contestants, etc. During the game, the other two players each went backstage to guess the rule and emerged with crowns.
Brennan is a calculating, competitive person (but able to laugh at himself) and at the end of the episode realized that the rule was essentially a prank on him, that he cannot win. He improvised this monologue to close out the show.
EDIT: Got some questions about where you can find this, and I'm a fan so I'm sharing! This is CollegeHumor, you might remember their YouTube videos like Jake & Amir from the last decade and a half. A couple years ago they started their own paid streaming service Dropout, so in addition to their normal sketches/clips on Youtube they began streaming a bunch of unscripted content like Game Changer (this clip), Um, Actually (a trivia show where contestants correct false statements about their favourite fandoms), and Dimension 20 (a wonderful D&D show hosted by Brennan Lee Mulligan, where every season is a new world he's created).
At the beginning of 2020 their holding company IAC stopped funding CollegeHumor and laid off virtually everyone. Sam Reich (this clip's host and the company's Chief Creative Officer) decided to buy CollegeHumor from IAC and kept Dropout going. They've still posted some content on YouTube during the pandemic, but mainly they've stayed the course for the last year with Dropout shows like the ones I mentioned above. I appreciate that having no clue who any of these people are. I watched this and my main parting thought was something like:
“Well that was clearly well produced. But, what the fuck did I just watch?”
I lost it at "I've solved your labyrinth, puzzle master. The minotaur has escaped and you're gonna to get the horns buddy."
That guy looks a lot like the CEO of Tide.

Brennan's oneshot DnD session with the McElroy brothers from The Adventure Zone and My, My Brother and Me is one of my favorite oneshots and DnD videos.
edit: For some reason Episode 2 & 3 got set to private, but I guess you can watch all 6 episodes on their streaming site. Kind of a bummer.
He also did Dadlands with them. So fucking funny
I don't know who these people are or what this show is, but I loved it. He reminded me of a young Jim Carey.
I've nothing of substance to add to this post other than the fact that I get to brag that I've had dinner with Brennan Lee Mulligan, and I've corresponded with him via email a couple of times. He's as swell as he seems, if not just a bit calmer in person.

I was one of the camera operators on this show and when Brennan started delivering this monologue, I knew it was going to be the best piece of improv I would ever experience first hand. I almost ruined the take by laughing so hard. I had to take my hands off the camera completely otherwise I would have knocked it over. One of the best shoots I have ever worked on in my career.

Dropout is probably my favorite streaming website. Game Changer, Umm Actually and Dimension 20 are worth every penny.
I signed up for Dropout for D20 and Game Changer, which like you said was already worth every penny. But then you take into account the back catalogues of stuff available! Paranoia was great, I don’t know WHY I waited so long to watch Total Forgiveness, and they have all of the old Jake and Amir sketches?? Solid gold. I agree with you, def my favorite streaming site
